- The immunogen corresponds to amino acids 346–484 of human GSDMD targeting the C‑terminal portion; therefore the antibody recognizes full‑length (~53 kDa) and the caspase‑cleaved C‑terminal fragment (~30 kDa) but not the N‑terminal pore‑forming domain.
- In Western blot, a dominant band at approximately 53 kDa is expected; upon inflammasome activation, a smaller band (~30 kDa) representing the C‑terminal cleavage product may also be visible.
- For immunofluorescence and immunohistochemistry, optimize fixation and permeabilization to detect GSDMD in its multiple subcellular locations (cytosol, membrane, mitochondria, nucleus).
