Biological Background: Gasdermin-D Function and Localization
- Gasdermin-D is a pore-forming protein precursor: upon cleavage by inflammatory caspases (CASP1, CASP4/CASP11), the N-terminal fragment oligomerizes to form membrane pores, triggering pyroptosis and the release of mature interleukin-1 (IL-1β and IL-18). - The N-terminal domain specifically binds to inner leaflet lipids, including monophosphorylated and bisphosphorylated phosphatidylinositols, phosphatidic acid, and cardiolipin, while it does not bind to unphosphorylated phosphatidylinositol, phosphatidylethanolamine, or phosphatidylcholine. Experimental Guidance and Technical Tips

- For western blot (WB), expect to detect full-length GSDMD at ~53 kDa and the cleaved N-terminal fragment at approximately 30–35 kDa upon inflammasome activation. Validate band patterns in your sample system with appropriate positive and negative controls.
- For immunofluorescence (IF/ICC), consider using inflammasome activators (e.g., LPS plus nigericin) to induce GSDMD cleavage and observe translocation from the cytosol to the plasma membrane. Co-staining with membrane or pyroptosis markers may help confirm pore formation.
- The antibody is suitable for ELISA, but optimal coating conditions and detection ranges should be empirically determined for each assay format.
