Biological Background: GSDMD Function and Localization
- Gasdermin-D (GSDMD) is a member of the gasdermin family and serves as a precursor of a pore-forming protein critical for pyroptosis and host defense. Related references: PMID:26375003, PMID:26375259, PMID:27281216
- Upon cleavage by inflammatory caspases (CASP1/4/5), the N-terminal fragment translocates to the plasma membrane, binds inner leaflet lipids such as phosphatidylinositol phosphates, and oligomerizes to form 10-15 nm pores, releasing IL-1β and IL-18 and triggering pyroptosis. Related references: PMID:27281216, PMID:27418190, PMID:29898893
- GSDMD pores also mediate mitochondrial membrane permeabilization, leading to mtDNA release, and the N-terminal fragment exhibits direct bactericidal activity against both Gram-negative and Gram-positive bacteria. Related references: PMID:27281216
- In the small intestine, a CASP3/7-cleaved fragment acts as a transcription coactivator for STAT1, promoting CIITA and MHC class II expression to maintain food tolerance.
- GSDMD is expressed in differentiating cells of the esophagus and stomach, and its activation in brain endothelial cells can cause blood-brain barrier breakdown during excessive pyroptosis.
- Post-translational modifications include palmitoylation, phosphorylation, and ubiquitination, and it belongs to the gasdermin family characterized by a conserved N-terminal pore-forming domain.
- Cleavage by papain allergen generates a pore-forming fragment that specifically releases IL-33, driving type 2 inflammation. Related references: PMID:35794369
Experimental Guidance and Technical Tips
- The immunogen (aa 346-484) corresponds to the C-terminal portion; therefore, this antibody may recognize both full-length GSDMD and the cleaved C-terminal fragment. For Western blot, consider validating detection of endogenous protein under inflammasome-activating conditions to distinguish cleaved species.
- A band at approximately 53 kDa represents full-length GSDMD; cleavage releases the N-terminal pore-forming domain (~31 kDa) and C-terminal fragment (~22 kDa). Use appropriate controls (e.g., LPS+ATP-treated macrophages) to confirm specificity.
- For ELISA, the antibody may be suitable as a capture or detection reagent. Consider pairing with an N-terminal-specific antibody to differentially quantify full-length versus cleaved GSDMD. Optimize coating and blocking conditions empirically.
- Since GSDMD forms oligomeric pores, solubility and conformation may vary across sample types; include mild detergents and sonication in lysis buffers for efficient extraction.
