Biological Background: Gasdermin-A Function and Localization
- Gasdermin-A (GSDMA), also known as Gasdermin-1, is a member of the gasdermin family of pore-forming proteins.
- It is expressed predominantly in the gastrointestinal tract and at lower levels in the skin and mammary gland; it is down-regulated in gastric cancer cells.
- GSDMA acts as a sensor of S.pyogenes infection: the bacterial protease SpeB cleaves GSDMA, releasing the N-terminal fragment.
- The N-terminal fragment binds to acidic phospholipids (e.g., cardiolipin, phosphatidylserine) on the inner leaflet of the cell membrane and oligomerizes, forming pores of 10–15 nm inner diameter. Related references: PMID:27281216, PMID:35110732, PMID:35545676

- Subcellularly, full-length GSDMA localizes to the cytoplasm and perinuclear region, while the cleaved N-terminal fragment translocates to the cell membrane.

Experimental Guidance and Technical Tips
- For Western blotting, consider that the antibody was raised against amino acids 61–181, and may detect both full-length (~49 kDa) and the cleaved N-terminal fragment in infected or stimulated samples. Validate with appropriate controls.
- For immunofluorescence (IF-P), note that GSDMA localizes to the cytoplasm and, upon cleavage, translocates to the membrane. Optimize fixation and permeabilization to preserve this localization.
- For ELISA, the antibody can be used to detect GSDMA in cell lysates or tissue homogenates; cross-reactivity with mouse allows use in mouse models.
- As the antibody cross-reacts with mouse, it is suitable for translational studies in mouse models of infection or cancer. Consider sequence alignment to confirm epitope conservation.
- As a rabbit monoclonal antibody, it offers high specificity and reproducibility; validate in the relevant sample system.
