Biological Background: GRSF1 Function and Localization
- G-rich sequence factor 1 (GRSF1) is a regulator of post-transcriptional mitochondrial gene expression, essential for mitochondrial ribosome assembly and the recruitment of mRNA and lncRNA (PubMed:23473033, PubMed:23473034). Related references: PMID:23473033, PMID:23473034
- It binds RNAs containing a 14-base G-rich element, with preference for ND6 mRNA and the long non-coding RNAs for MT-CYB and MT-ND5.
- GRSF1 facilitates the degradosome-mediated decay of non-coding mitochondrial transcripts by unwinding G-quadruplex (G4) RNA structures (PubMed:29967381). Related references: PMID:29967381
- The protein localizes to the mitochondrial matrix and can also be found in the cytoplasm.
- GRSF1 contains RNA-binding domains and is a phosphoprotein; alternative splicing produces multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 50–150 of human GRSF1, a region well conserved across human, mouse, and rat. The antibody is expected to recognize all major isoforms.
- For Western blot, the predicted molecular weight is ~53 kDa. Validate using mitochondrial-enriched lysates from relevant cell lines or tissues.
- In immunofluorescence/ICC, GRSF1 exhibits mitochondrial localization. Co-staining with mitochondrial markers (e.g., TOM20) is recommended to confirm specificity.
- For immunoprecipitation, the antibody can be used to enrich endogenous GRSF1 and associated RNA complexes. Include appropriate controls.
- ELISA applications may be useful for quantitative detection; optimal coating and detection conditions should be determined empirically.
