Biological Background: Grp94 Function and Localization
- Endoplasmin (Grp94, also known as gp96, TRA1) is a 92 kDa glucose-regulated protein and a member of the Hsp90 family, functioning as an ATP-dependent molecular chaperone in the endoplasmic reticulum.
- It plays a critical role in protein folding and trafficking, including the proper folding of Toll-like receptors (TLRs) such as TLR4, in cooperation with CNPY3, ensuring their cell surface expression and innate immune function. Related references: PMID:11584270
- Grp94, together with MESD, modulates the canonical Wnt signaling pathway by promoting the folding of the coreceptor LRP6, thus influencing cell differentiation and development. Related references: PMID:23572575, PMID:39509507
- It also facilitates the unconventional secretion of leaderless cargos such as interleukin-1 (IL-1) by unfolding them and enabling TMED10-mediated translocation into the ERGIC for secretion. Related references: PMID:32272059
- Subcellularly, Grp94 resides in the lumen of the endoplasmic reticulum and sarcoplasmic reticulum, and is also found in melanosomes, reflecting its role in protein processing and transport.
- The protein undergoes various post-translational modifications including acetylation, hydroxylation, phosphorylation, and disulfide bond formation, and contains an ATP-binding domain essential for its chaperone activity.
- As a high-abundance ER resident, Grp94 is implicated in cellular stress responses and has been identified as a tumor rejection antigen, highlighting its importance in cancer immunology.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a sequence within amino acids 40–310 of human Grp94, targeting the N-terminal region; consider this when designing experiments with truncated or mutated proteins.
- For Western blot, a band at approximately 92 kDa is expected; validate the antibody in your specific sample system to confirm reactivity.
- This antibody has been reported to work in WB, IF/ICC, IF-P, IHC-P, and ELISA; for each application, optimization of dilution and protocol is recommended.
- Cross-reactivity is documented for human, mouse, and rat; however, due to high homology among Hsp90 family members, confirm specificity for your target in relevant species.
- Store the antibody as recommended (shipped on ice) and avoid repeated freeze-thaw cycles to maintain activity.
