Biological Background: GRO alpha Function and Localization
- Growth-regulated alpha protein, also known as C-X-C motif chemokine 1 (CXCL1), melanoma growth stimulatory activity (MGSA), and neutrophil-activating protein 3 (NAP-3).
- Encoded by the CXCL1 gene (GeneID: 2919) in humans.
- It is a small, secreted cytokine belonging to the CXC chemokine family, characterized by conserved cysteine residues forming disulfide bonds (UniProt keyword: Disulfide bond).
- Exhibits potent chemotactic activity for neutrophils, playing a critical role in inflammatory responses (UniProt keyword: Inflammatory response, Cytokine).
- In vitro, processed forms GRO-alpha(4-73), GRO-alpha(5-73) and GRO-alpha(6-73) show a 30-fold higher chemotactic activity compared to the full-length protein.
- May function in an autocrine manner on endothelial cells, suggesting involvement in angiogenesis and wound healing.
- Subcellular localization is predominantly secreted (UniProt: Secreted).
- Post-translational modifications include signal peptide cleavage and disulfide bond formation (UniProt keyword: Signal, Disulfide bond).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide covering amino acids 1-107, which includes the signal peptide; this may allow detection of both precursor and mature forms.
- For Western blot, consider reducing conditions to resolve the 11 kDa band effectively, given the presence of disulfide bonds.
- In immunofluorescence/ICC, note that GRO alpha is secreted; optimize fixation and permeabilization to retain the target.
- ELISA application may require pairing with a suitable detection antibody; validate coating conditions and standard curves in your assay.
- Cross-reactivity is reported for human and rat; reactivity with other species should be verified individually.
