Biological Background: G Protein-Coupled Receptor Kinase 3 Function and Localization

  • GRK3, also known as Beta-adrenergic receptor kinase 2 (Beta-ARK-2) or BARK2, is a serine/threonine-protein kinase belonging to the G protein-coupled receptor kinase family.
  • It specifically phosphorylates activated G protein-coupled receptors (GPCRs), triggering beta-arrestin-mediated receptor internalization and desensitization. Related references: PMID:21799898
  • GRK3 phosphorylates ligand-bound anaphylatoxin receptors C3AR1 and C5AR1, contributing to their desensitization, and also acts on beta-adrenergic and related receptors by similarity. Related references: PMID:21799898
  • It positively regulates the ciliary smoothened (SMO) signaling pathway by phosphorylating SMO, which promotes SMO ciliary trafficking and activity (by similarity).
  • Localized to both presynaptic and postsynaptic compartments, GRK3 is involved in neuronal signal modulation.
  • The protein features an ATP-binding domain and is subject to ubiquitination, reflecting its regulatory roles.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to the C-terminal region (residues 589–688) of human GRK3, favoring specificity for the target.
  • For Western blotting, the predicted molecular weight is approximately 80 kDa; consider using appropriate positive controls from human cell lines or tissues.
  • In ELISA applications, validate antibody performance with the relevant antigen and experimental conditions.
  • Confirm cross-reactivity in the intended human sample system, as the antibody is not tested in other species.
