Biological Background: G Protein-Coupled Receptor Kinase 3 Function and Localization
- GRK3, also known as Beta-adrenergic receptor kinase 2 (Beta-ARK-2) or BARK2, is a serine/threonine-protein kinase belonging to the G protein-coupled receptor kinase family.
- It specifically phosphorylates activated G protein-coupled receptors (GPCRs), triggering beta-arrestin-mediated receptor internalization and desensitization. Related references: PMID:21799898
- GRK3 phosphorylates ligand-bound anaphylatoxin receptors C3AR1 and C5AR1, contributing to their desensitization, and also acts on beta-adrenergic and related receptors by similarity. Related references: PMID:21799898
- It positively regulates the ciliary smoothened (SMO) signaling pathway by phosphorylating SMO, which promotes SMO ciliary trafficking and activity (by similarity).
