Biological Background: GRK2 Function and Localization
- GRK2 (G protein-coupled receptor kinase 2), also known as beta-adrenergic receptor kinase 1 (Beta-ARK-1), is a serine/threonine-protein kinase that specifically phosphorylates activated forms of G protein-coupled receptors (GPCRs).
Related references: PMID:14976224, PMID:16179383, PMID:19306925 - Receptor phosphorylation by GRK2 initiates beta-arrestin-mediated receptor internalization and desensitization, thereby attenuating GPCR signaling.
Related references: PMID:14976224, PMID:16179383, PMID:19306925 - GRK2 phosphorylates a variety of GPCRs including the beta-2 adrenergic receptor (ADRB2), C3a and C5a anaphylatoxin receptors (C3AR1 and C5AR1), lysophosphatidic acid receptors (LPAR1 and LPAR2), smoothened (SMO), and GPR161, modulating their signaling and trafficking.
Related references: PMID:19715378, PMID:21799898, PMID:23077507, PMID:39138140, PMID:40384633, PMID:19306925, PMID:30284927 - GRK2 localizes to the cytoplasm, cell membrane, and cilium membrane, and is also found at postsynaptic and presynaptic compartments.
- As a phosphoprotein and ATP-binding kinase, GRK2 belongs to the serine/threonine protein kinase family and serves as a key regulator of cellular signaling dynamics.
- GRK2 is expressed in peripheral blood leukocytes, underscoring its role in immune and inflammatory responses.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 440–689 of human GRK2, targeting the C-terminal region; consider that this antibody may recognize both full-length GRK2 and potential C-terminal truncation variants.
- For Western blotting (WB), a band at approximately 80 kDa is expected; validate detection in your sample system and consider using appropriate positive and negative controls.
- In immunohistochemistry (IHC-P), optimize fixation and antigen retrieval conditions; test a range of antibody dilutions on control tissues.
- For ELISA applications, the antibody can be used as a capture or detection reagent; perform cross-titration experiments to determine optimal working concentrations.
