Biological Background: GRIK2 Function and Localization

  • GRIK2, also known as Glutamate receptor ionotropic, kainate 2 (GluK2, GluR6, EAA4), is a member of the kainate family of ionotropic glutamate receptors, encoded by the GRIK2 gene in human.
  • It functions as a cation-permeable ligand-gated ion channel, activated by the neurotransmitter L-glutamate and the agonist kainic acid, mediating fast excitatory synaptic transmission in the central nervous system. Related references: PMID:7536611 PMID:8730589 PMID:14511640
  • Upon agonist binding, the receptor opens its channel and then rapidly desensitizes, entering a transient inactive state characterized by bound agonist. Related references: PMID:14511640 PMID:28180184 PMID:34375587
  • In association with GRIK3, GRIK2 is involved in presynaptic facilitation of glutamate release at hippocampal mossy fiber synapses.
  • In addition to its ionotropic role, GRIK2 acts as a thermoreceptor conferring sensitivity to cold temperatures, particularly in dorsal root ganglion neurons. Related references: PMID:31474366
  • GRIK2 is primarily localized to the cell membrane, specifically at the postsynaptic membrane of excitatory synapses.
  • Expression is higher in the cerebellum compared to the cerebral cortex, reflecting its role in specific neural circuits.
  • The receptor undergoes post-translational modifications including phosphorylation, glycosylation, and disulfide bond formation, which regulate its function and trafficking.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal extracellular domain (amino acids 30-300) of human GRIK2, which may help in recognizing both native and denatured receptor forms; consider using a mild denaturing gel for WB to preserve epitopes if expected bands are absent.
  • For western blotting, a protein band at approximately 103 kDa is expected; standard protocols for high-molecular-weight membrane proteins (e.g., loading at 4°C, using 5-8% SDS-PAGE) are recommended.
  • The antibody cross-reacts with both human and mouse GRIK2; validate the reactivity in your specific sample type (e.g., brain lysate, transfected cells) with appropriate positive and negative controls.
  • For ELISA applications, the polyclonal nature may provide enhanced sensitivity; however, lot-to-lot consistency should be verified for quantitative assays.
