Biological Background: GRID2 Function and Localization
- GRID2 encodes glutamate receptor ionotropic delta-2 (GluD2), a member of the ionotropic glutamate receptor family structurally similar to other iGluR subunits but atypical in ligand binding.
- Although it shares the classical iGluR topology, GluD2 does not bind glutamate as its primary ligand (PubMed:34936451). Instead, it participates in non-canonical signaling complexes. Related references: PMID:34936451
- Core function: promotes synaptogenesis and mediates D-serine-dependent long-term depression (LTD) and AMPA receptor endocytosis at cerebellar parallel fiber–Purkinje cell synapses via the NRX1B–CBLN1–GRID2 triad complex (PubMed:27418511). Related references: PMID:27418511
- In the presence of neurexins and cerebellins, GluD2 forms cation-selective ion channels gated by glycine and D-serine (PubMed:34936451). Related references: PMID:34936451
- Ion channel activity can be triggered by metabotropic glutamate receptor 1 (GRM1) in Purkinje cells (PubMed:24357660, PubMed:27276689). Related references: PMID:24357660 PMID:27276689
- Subcellular localization: primarily at postsynaptic cell membranes of excitatory synapses, particularly enriched in the cerebellum.
- Features multiple post-translational modifications and structural motifs, including disulfide bonds, glycosylation, phosphorylation, and metal-binding sites, as reflected in Swiss-Prot keywords (alternative splicing, calcium, cell membrane, ion channel, ligand-gated ion channel, neurodegeneration, postsynaptic cell membrane, synapse, transmembrane).
Experimental Guidance and Technical Tips
- Immunogen design: the synthetic peptide spans amino acids 900–1007 in the intracellular C-terminal region; the antibody may recognize full-length and potential C-terminal fragments.
- Western blotting: a band near 113 kDa is expected; use cerebellar or brain lysates as positive controls. Validate with siRNA knockdown or CRISPR knockout samples if possible.
- IF/ICC: permeabilization is recommended to access the cytoplasmic epitope. Staining should localize to synaptic puncta in neuronal cultures.
- ELISA: suitable for quantitative detection; consider pre‑incubation with the immunizing peptide to verify signal specificity.
- Cross-reactivity: predicted to react with human, mouse, and rat based on sequence homology; perform your own verification in the experimental system.
