Biological Background: GREM1 (Gremlin-1) Function and Localization

  • Gremlin-1 is a secreted cytokine belonging to the DAN domain family, also known as cysteine knot superfamily 1, BMP antagonist 1, and cell proliferation-inducing gene 2 protein. It plays critical roles in development and disease through bone morphogenetic protein (BMP) antagonism.
  • Functions as a potent BMP antagonist, essential for early limb outgrowth and patterning by maintaining the FGF4-SHH feedback loop. It down-regulates BMP4 signaling in a dose-dependent manner (By similarity).
  • Antagonizes BMP2, inhibiting BMP2-mediated differentiation of osteoblasts in vitro. Related references: PMID:27036124
  • Acts as an inhibitor of monocyte chemotaxis and may influence carcinogenesis and metanephric kidney organogenesis (By similarity).
  • Overexpression can inhibit the growth or viability of normal cells but not transformed cells (By similarity).
  • Secreted into the extracellular space, where it interacts with BMP ligands to modulate signaling.
  • Highly expressed in the small intestine, fetal brain, and colon. In the colon, expression is normally restricted to intestinal subepithelial myofibroblasts at the crypt base, but in HMPS1 subjects, it is also strongly expressed in epithelial cells along the crypt.
  • The protein undergoes alternative splicing and is glycosylated, with a predicted molecular weight of ~21 kDa. It contains disulfide bonds characteristic of the cystine knot superfamily.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 44–143 of human GREM1, covering a central region of the protein. The antibody is expected to recognize both wild-type and splice variants sharing this sequence.
  • For Western blotting, GREM1 runs at approximately 21 kDa under reducing conditions; however, glycosylation may cause a shift to a higher apparent molecular weight. Validate detection in relevant cell lysates or tissue extracts.
  • ELISA applications may benefit from using recombinant GREM1 as a standard, but cross-reactivity with other BMP antagonists should be assessed experimentally.
  • Tissue specificity suggests strong expression in small intestine, colon, and fetal brain. Consider these as positive controls and validate in your model system.
  • Since GREM1 is secreted, cell culture supernatants can be used for detection after concentration if needed.
