Biological Background: GRASP65 (GORASP1) Function and Localization
- GRASP65 (GORASP1, GOLPH5) is a key structural protein of the Golgi apparatus, belonging to the GORASP family. Alternative splicing produces multiple isoforms. (Keywords: Alternative splicing)
- It is required for the formation and maintenance of the Golgi ribbon, acting in concert with GORASP2/GRASP55. It also plays a role in assembly and membrane stacking of cisternae, and in the reassembly of Golgi stacks after mitotic breakdown.
Related references:
PMID:33301566 - Caspase-mediated cleavage of GRASP65 is necessary for Golgi fragmentation during apoptosis. (By similarity)
- GRASP65 mediates, via interaction with GOLGA2/GM130, the docking of transport vesicles with Golgi membranes.
Related references:
PMID:16489344 - It facilitates ER stress-induced unconventional trafficking of core-glycosylated CFTR to the cell membrane.
Related references:
PMID:21884936 - Subcellularly, GRASP65 localizes to the Golgi apparatus, specifically the cis-Golgi network membrane and the endoplasmic reticulum-Golgi intermediate compartment membrane.
- Post-translational modifications include phosphorylation, myristoylation, and zinc-binding. The protein is a phosphoprotein, lipoprotein, and metal-binding protein.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 221–440 of human GRASP65, located in the C-terminal region. When predicting isoform recognition, consider potential splice variants affecting this domain.
- For Western blotting (WB), validate the antibody in human cell lines; endogenous GRASP65 migrates at approximately 46 kDa. Cleavage fragments may appear during apoptosis—use appropriate controls (e.g., staurosporine-treated cells).
- In immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation (e.g., methanol or paraformaldehyde) and permeabilization to achieve clear Golgi staining. Co-staining with a Golgi marker (e.g., GM130) is recommended.
- For ELISA, the antibody can be used as a capture or detection reagent; titrate to determine optimal concentration for your assay system.
