Biological Background: GRASP65 Function and Localization
- GRASP65, encoded by the GORASP1 gene, is also known as Golgi peripheral membrane protein p65 or GOLPH5.
- It serves as a key structural protein of the Golgi apparatus, cooperating with GORASP2/GRASP55 to form and maintain the Golgi ribbon. Related references: PMID:33301566
- GRASP65 participates in the assembly and membrane stacking of Golgi cisternae and is critical for the reassembly of Golgi stacks after mitotic breakdown.
- During apoptosis, caspase-mediated cleavage of GRASP65 triggers Golgi fragmentation.
- Through its interaction with GOLGA2/GM130, GRASP65 mediates the docking of transport vesicles at Golgi membranes. Related references: PMID:16489344
- It facilitates ER stress-induced unconventional trafficking of core-glycosylated CFTR to the plasma membrane. Related references: PMID:21884936
- Subcellularly, GRASP65 localizes to the Golgi apparatus, primarily at the cis-Golgi network and ER-Golgi intermediate compartment membranes.
- Post-translational modifications include phosphorylation and myristoylation, reflecting its regulation and membrane association.
Experimental Guidance and Technical Tips
- For Western blotting, expect a band near 46 kDa; validate with appropriate molecular weight markers.
- In immunofluorescence, the antibody should exhibit a Golgi-like staining pattern; optimize fixation and permeabilization conditions.
- The immunogen (aa 341–440) lies in the C-terminal region; cleavage fragments may not be detected; consider using full-length controls.
- For ELISA applications, titrate the antibody to determine optimal coating or detection concentrations.
- Cross-reactivity with human, mouse, and rat has been reported; verify performance in the species of interest.
