Biological Background: Granzyme B Function and Localization

  • Granzyme B (CTLA-1, CCP1, Fragmentin-2) is a serine protease encoded by the Gzmb gene, functioning as a key effector in immune cell-mediated cytotoxicity.
  • It is stored in cytolytic granules of cytotoxic T-cells and natural killer (NK) cells and secreted upon immune synapse formation.

  • It can also trigger pyroptosis via cleavage of gasdermin-E (GSDME), releasing the pore-forming moiety.
