Biological Background: Granzyme B Function and Localization
- Granzyme B (CTLA-1, CCP1, Fragmentin-2) is a serine protease encoded by the Gzmb gene, functioning as a key effector in immune cell-mediated cytotoxicity.
- It is stored in cytolytic granules of cytotoxic T-cells and natural killer (NK) cells and secreted upon immune synapse formation.

- It can also trigger pyroptosis via cleavage of gasdermin-E (GSDME), releasing the pore-forming moiety.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 141–240; therefore, the antibody may recognize both full-length and cleaved forms of Granzyme B.
- For Western blot, a band at approximately 27 kDa is expected in mouse cytotoxic T-cell or NK-cell lysates. Validate with appropriate controls.
- For ELISA, consider using the immunizing peptide as a positive control and test serial dilutions of the antibody.
- Since this is a polyclonal antibody, lot-to-lot variation may occur; include positive and negative controls in every experiment.
- Cross-reactivity with other species is not expected; confirm specificity using mouse Granzyme B knockout samples if available.
