Biological Background: Granulin Function and Localization

  • Progranulin (GRN, also known as GEP, GP88, PEPI, PGRN, CLN11, PCDGF) is a secreted glycoprotein critical for lysosomal homeostasis, regulating protein trafficking to lysosomes, lysosomal enzyme activity, and acidification, which facilitates CTSB-mediated degradation of mature CTSD. Related references: PMID:28453791, PMID:28541286, PMID:28073925
  • It acts as a growth factor in wound healing and inflammation, directly promoting dermal fibroblast and endothelial cell division, migration, and capillary-like tubule formation, and also protects epithelial cells by blocking TNF-mediated neutrophil activation. Related references: PMID:12526812
  • Progranulin modulates neuronal inflammation, supporting neuronal survival, axonal outgrowth, and integrity, with implications in neurodegeneration. Related references: PMID:18378771
  • Proteolytic processing of the full-length protein generates various granulin peptides, some of which independently inhibit epithelial cell proliferation and induce IL-8 secretion.
  • Progranulin also stabilizes cathepsin D (CTSD) through direct interaction, maintaining its aspartic-type peptidase activity.
  • The protein is localized both extracellularly (secreted) and within lysosomes, consistent with its roles in growth factor signaling and lysosomal function.
  • It is widely expressed, with highest levels in the kidney, and is found in epithelial cells, fibroblasts, myelogenous leukemic cell lines, inflammatory cells, and bone marrow.
  • The progranulin protein sequence contains multiple granulin repeats stabilized by disulfide bonds, and it undergoes glycosylation. Keywords: Alternative splicing, Disulfide bond, Glycoprotein, Signal, Repeat.

Experimental Guidance and Technical Tips

  • The immunogen (aa 18-593) covers the mature progranulin protein, so this polyclonal antibody may detect full-length progranulin (~64 kDa) and potentially some processed granulin peptides in WB.
  • For Western blotting, use a reducing SDS-PAGE and validate the expected band size; glycosylation may cause slight shifts. Include appropriate positive controls such as epithelial cell lines or kidney tissue lysates.
  • For ELISA, consider pairing with a monoclonal capture antibody to enhance specificity, and validate detection in serum or cell culture supernatants where progranulin is secreted.
  • Cross-reactivity with mouse and rat is reported; however, users should validate the antibody in their specific experimental system to confirm reactivity and optimal dilution.
  • Since progranulin is involved in lysosomal function, consider using lysosomal inhibitors in cell-based assays to study its processing and trafficking.
