Biological Background: Granulin Function and Localization
- Progranulin (GRN), also known as GP88, PCDGF, proepithelin, and acrogranin, is a secreted glycoprotein belonging to the granulin family.
- Acts as a key regulator of lysosomal function and a growth factor involved in inflammation, wound healing, and cell proliferation (PubMed:12526812, 18378771, 28073925). Related references: PMID:12526812, PMID:18378771, PMID:28073925
- Regulates protein trafficking to lysosomes and the activity of lysosomal enzymes (PubMed:28453791, 28541286). Related references: PMID:28453791, PMID:28541286
- Facilitates lysosomal acidification, causing degradation of mature CTSD by CTSB (PubMed:28073925). Related references: PMID:28073925
- Promotes epithelial cell proliferation by blocking TNF-mediated neutrophil activation, preventing release of oxidants and proteases (PubMed:12526812). Related references: PMID:12526812
- Modulates inflammation in neurons by preserving neuron survival, axonal outgrowth, and neuronal integrity (PubMed:18378771). Related references: PMID:18378771
- Additionally, it can inhibit epithelial proliferation and induce IL-8 secretion, and it stabilizes CTSD to maintain its aspartic-type peptidase activity.
- Subcellularly localizes to the lysosome and is secreted; highest tissue expression is observed in kidney, with presence in inflammatory cells and various leukemic cell lines.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal region of human Progranulin; consider sequence alignment to assess cross-reactivity with isoforms or orthologs.
- For Western blot, the predicted molecular weight of full-length Progranulin is ~64 kDa, but cleavage products may be observed; optimize sample preparation and validate with appropriate positive controls.
- For ELISA, validate coating concentration and detection conditions in your specific sample matrix to ensure optimal signal-to-noise ratios.
- Cross-reactivity is reported with human and mouse; when using mouse samples, confirm specificity with appropriate controls due to potential endogenous IgG interference in certain assay formats.
