Biological Background: GPX4 Function and Localization
- GPX4 is a phospholipid hydroperoxide glutathione peroxidase that directly reduces phospholipid, cholesterol, and thymine hydroperoxides, even when incorporated into membranes and lipoproteins. Related references: PMID:40281343
- It plays a critical role in preventing ferroptosis and protecting cells from oxidative damage by eliminating lipid reactive oxygen species. Related references: PMID:24439385 PMID:40281343
- The active site selenocysteine is essential for oxidative resistance, neuronal survival, and ferroptosis prevention, with Cys substitution resulting in lethality (By similarity).
- Localizes to both mitochondria and cytoplasm, enabling antioxidant function in multiple subcellular compartments.
- Expressed primarily in testis and found in platelets at the protein level. Related references: PMID:11115402
- Involved in arachidonic acid metabolism in platelets and reduces fatty acid-derived hydroperoxides. Related references: PMID:11115402
- Also reduces small soluble hydroperoxides such as H2O2, cumene hydroperoxide, and tert-butyl hydroperoxide. Related references: PMID:17630701 PMID:36608588
- Required for sperm development, photoreceptor cell maturation, and T-cell expansion during immune response (By similarity).
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 74–197 of human GPX4, encompassing the active site selenocysteine region; this design may recognize both mitochondrial and cytoplasmic isoforms.
- For Western blotting, expect bands around 20 kDa and 22 kDa due to alternative initiation or post-translational modifications; validate size shifts using relevant controls.
- In immunofluorescence and immunohistochemistry, consider mitochondrial and cytoplasmic staining patterns; co-localization with organelle markers is recommended for confirmation.
- ELISA performance may vary; titrate antibody and use appropriate blocking to minimize background, especially with platelet or testis lysates.
- Cross-reactivity with mouse and rat GPX4 is supported; however, verify sequence homology in the epitope region for your species of interest.
