Biological Background: GPVI/GP6 Function and Localization

- It is critical for platelet procoagulant activity, driving thrombin generation and fibrin formation, which contributes to thrombus development in arteries and veins.
- Downstream signaling is transduced via association with the FcR gamma-chain, involving Src family kinases (LYN or FYN), SYK, and adapter protein LAT, ultimately activating PLCG2.
- GPVI localizes to the cell membrane and is expressed exclusively in megakaryocytes and platelets.

- The immunogen corresponds to the extracellular domain (aa 24-265) of mouse GPVI; this region may share sufficient homology with human GPVI to enable the observed cross-reactivity, but validation in human platelet samples is recommended.
- For Western blotting, the predicted molecular weight is ~34 kDa; glycosylation and other post-translational modifications may cause a shift in apparent molecular weight; consider using platelet lysates and appropriate controls.
- In ELISA applications, the antibody may be used for direct detection of recombinant GPVI or for capture/detection in sandwich assays; optimize coating and blocking conditions using mouse or human platelet-derived material.
