Biological Background: GPRC6A Function and Localization

• GPRC6A is a G protein-coupled receptor of the class C family, also designated as GPCR33, encoded by the gene GPRC6A.

• Functions as a promiscuous receptor activated by diverse ligands, including osteocalcin (BGLAP), basic amino acids (L-Lys, L-Arg, L-ornithine), small polar amino acids, and divalent cations such as Ca²⁺ and Mg²⁺. Related references: PMID:15576628

• Signals primarily through G(q)/G(11) and G(i)-coupled intracellular pathways.

• In testicular Leydig cells, mediates osteocalcin-dependent upregulation of steroidogenic enzymes, promoting testosterone biosynthesis via CREB activation.
