Biological Background: GPRC6A Function and Localization
-
GPRC6A is a G protein-coupled receptor of the class C family, also designated as GPCR33, encoded by the gene GPRC6A.
-
Functions as a promiscuous receptor activated by diverse ligands, including osteocalcin (BGLAP), basic amino acids (L-Lys, L-Arg, L-ornithine), small polar amino acids, and divalent cations such as Ca²⁺ and Mg²⁺. Related references: PMID:15576628
-
Signals primarily through G(q)/G(11) and G(i)-coupled intracellular pathways.
-
In testicular Leydig cells, mediates osteocalcin-dependent upregulation of steroidogenic enzymes, promoting testosterone biosynthesis via CREB activation.
-
Mediates non-genomic effects of androgens in multiple tissues.
-
Integrates nutritional and hormonal anabolic signals by sensing extracellular amino acids, osteocalcin, divalent ions, and anabolic steroids. Related references: PMID:20947496
-
Localizes to the cell membrane and possesses characteristic structural features including a signal peptide, seven transmembrane domains, disulfide bonds, and N-linked glycosylation sites.
-
Isoform 1 shows high expression in brain, skeletal muscle, testis, bone, calvaria, osteoblasts, and leukocytes; moderate in liver, heart, kidney, and spleen; low in lung, pancreas, placenta, and ovary. Isoform 2 is expressed at lower levels except in kidney where it equals isoform 1.
Experimental Guidance and Technical Tips
- The immunogen (aa 300–590 of human GPRC6A) corresponds to a region containing both extracellular and transmembrane domains; polyclonal antibodies may detect multiple epitopes. Validate specificity with appropriate controls.
- For Western blot, use tissue lysates with known GPRC6A expression (e.g., brain, testis, skeletal muscle) and expect a band near 105 kDa; glycosylation may cause slight shifts.
- Optimize blocking and antibody dilutions empirically; the antibody is suitable for WB and ELISA.
- Cross-reactivity with mouse and rat allows multi-species studies; however, confirm detection in the relevant model system.
