Biological Background: GPRC5B Function and Localization
- GPRC5B (G protein-coupled receptor family C group 5 member B), also known as retinoic acid-induced gene 2 protein (RAIG-2) or A-69G12.1, is a member of the G protein-coupled receptor family C group 5.
- It functions as a receptor involved in the regulation of cell volume.
- The protein is localized to the cell membrane and cytoplasmic vesicle membranes.
- Tissue expression: High in kidney, pancreas, testis; medium in brain, heart, prostate, small intestine, spleen; low in liver, placenta, skeletal muscle, colon, ovary, thymus; not detected in lung and peripheral leukocyte. In the central nervous system, highest expression is observed in corpus callosum, caudate nucleus, putamen, substantia nigra, thalamus, hippocampus, spinal cord, and dorsal root ganglia (PubMed:10945465). It is expressed in glia limitans, ependymal cells, astrocytes, and perivascular regions, but not in neurons (PubMed:37143309). Related references: PMID:10945465 PMID:37143309
- The protein undergoes alternative splicing and contains a signal peptide, seven transmembrane helices, and undergoes glycosylation and phosphorylation.
- It is associated with disease variants.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide corresponding to amino acids 304-403 of human GPRC5B. For ELISA applications, consider using this peptide as a positive control or coating antigen.
- For Western blot, the predicted molecular weight is approximately 45 kDa. Validate band sizes in relevant cell or tissue lysates; expression data suggest kidney, pancreas, testis, and brain as positive sample sources.
- Cross-reactivity with mouse and rat indicates potential for multi-species detection, but verify performance in the species of interest.
- Optimize dilution and detection conditions empirically in your specific assay system.
