Biological Background: GPR62 Function and Localization
- GPR62, also known as GPCR8 or KPG_005, is an orphan G protein-coupled receptor encoded by the GPR62 gene (GeneID: 118442) in humans.
- It constitutively activates the G(q/11)/inositol phosphate and the G(s)-alpha/cAMP signaling pathways, and spontaneously recruits beta-arrestin. It also shows reciprocal modulation with the melatonin receptor MTNR1B, likely through receptor heteromerization. Related references: PMID:28827538
- Subcellularly, GPR62 localizes to the cell membrane and endosome membrane.
- Expression is mainly observed in the brain, with detection in the basal forebrain, frontal cortex, caudate, putamen, thalamus, and hippocampus.
- As a GPCR, it possesses transmembrane helices and is predicted to be glycosylated (glycoprotein), which may influence its trafficking and function.

Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic peptide (aa 150-250 of human GPR62, NP_543141.3). In Western blot, a band near 38 kDa is expected; validate in control brain tissue lysates from human, mouse, or rat.
- For ELISA, consider using the immunogen peptide as a positive control to check assay sensitivity and specificity.
- Since the epitope is linear, denaturing and reducing conditions in SDS-PAGE may expose the target epitope. Optimization of sample preparation and antibody dilution is recommended for both WB and ELISA.
- Cross-reactivity is reported for human, mouse, and rat, but testing in the desired species and tissue is advised to confirm performance.
