Biological Background: Prosaposin Receptor GPR37 Function and Localization

  • GPR37 is a G protein-coupled receptor that serves as a receptor for prosaposin, osteocalcin, and neuroprotectin D1 (NPD1). Ligand binding induces receptor endocytosis and ERK phosphorylation, mediating physiological processes such as inflammatory pain resolution and oligodendrocyte differentiation. Related references: PMID:11439185 PMID:23690594
  • It negatively regulates oligodendrocyte differentiation and central nervous system myelination during development through the ERK1/2 signaling pathway, and is thought to influence myelin stability and resistance to demyelination.
  • Activation by NPD1 promotes macrophage phagocytosis and shifts cytokine release toward an anti-inflammatory profile, contributing to the resolution of inflammatory pain and protection against sepsis.
  • GPR37 may act as a maturation factor for LRP6, protecting it from endoplasmic reticulum-associated protein degradation (ERAD) and thereby enhancing Wnt/β-catenin signaling. Related references: PMID:28341812
  • Subcellularly, the receptor localizes to the cell membrane, endoplasmic reticulum membrane, dendrites, and synapses, consistent with its roles in surface signaling and intracellular trafficking.
  • It is predominantly expressed in brain and spinal cord, with lower levels in testis, placenta, and liver. Overexpression leads to protein aggregation and is linked to dopaminergic neuronal death in juvenile Parkinson’s disease.
  • The protein is a multi-pass membrane glycoprotein with disulfide bonds, and is subject to ubiquitination. Keywords include G protein-coupled receptor, transmembrane helix, glycoprotein, and synapse.

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal extracellular region (amino acids 40–180), which is likely accessible in native receptor conformations; this antibody is suitable for both denaturing Western blot and native ELISA applications.
  • For Western blot, a 67 kDa band should be detected; use a 10% SDS-PAGE gel and lysates from GPR37-expressing tissues (e.g., brain) or overexpressing cells. Validate with proper controls due to possible aggregation.
  • In ELISA, the recombinant immunogen can serve as a positive control for coating and detection optimization.
  • This antibody cross-reacts with human and rat GPR37. For other species, check sequence homology of the immunogen region before use.
  • GPR37 is prone to aggregation upon overexpression; consider mild solubilization conditions to minimize aggregation artifacts in sample preparation.
