Biological Background: Prosaposin Receptor GPR37 Function and Localization
- GPR37 is a G protein-coupled receptor that serves as a receptor for prosaposin, osteocalcin, and neuroprotectin D1 (NPD1). Ligand binding induces receptor endocytosis and ERK phosphorylation, mediating physiological processes such as inflammatory pain resolution and oligodendrocyte differentiation. - It negatively regulates oligodendrocyte differentiation and central nervous system myelination during development through the ERK1/2 signaling pathway, and is thought to influence myelin stability and resistance to demyelination.
- Activation by NPD1 promotes macrophage phagocytosis and shifts cytokine release toward an anti-inflammatory profile, contributing to the resolution of inflammatory pain and protection against sepsis.

- For Western blot, a 67 kDa band should be detected; use a 10% SDS-PAGE gel and lysates from GPR37-expressing tissues (e.g., brain) or overexpressing cells. Validate with proper controls due to possible aggregation.
- In ELISA, the recombinant immunogen can serve as a positive control for coating and detection optimization.
- This antibody cross-reacts with human and rat GPR37. For other species, check sequence homology of the immunogen region before use.
