- GPR155 (Lysosomal cholesterol signaling protein, also known as PGR22, DEPDC3) functions as a cholesterol sensor that couples lysosomal cholesterol levels to mTORC1 signaling. In cholesterol-replete conditions, GPR155/LYCHOS binds cholesterol, disrupting the GATOR1-KICSTOR complex to promote mTORC1 activation; during cholesterol starvation, the GATOR1-KICSTOR interaction is maintained, inhibiting mTORC1.
