Biological Background: GP2 Function and Localization

  • GP2, also known as ZAP75, is a pancreatic secretory granule membrane major glycoprotein.
  • It functions as an intestinal M-cell transcytotic receptor specific for type-I-piliated bacteria, binding fimH protein on bacterial type I pilus tips.
  • Upon binding, GP2 internalizes bacteria such as E. coli and S. typhimurium and delivers them across M cells to lymphoid follicles, initiating mucosal immune responses.
  • GP2 is localized at the zymogen granule membrane, but also found secreted, at the cell membrane, apical membrane, membrane rafts, and endosomes.
  • GP2 is specifically expressed in pancreatic tissue (at the protein level). Related references: PMID:10760606 PMID:8666297
  • It is also specifically expressed by intestinal M (microfold) cells. Related references: PMID:19907495
  • Key features include a GPI-anchor, disulfide bonds, an EGF-like domain, and glycosylation; the protein exists in alternatively spliced forms.
  • The mature protein has a molecular weight of approximately 59 kDa (UniProt: 59480 Da).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a recombinant fragment (aa 111–387) of human GP2; consider that the antibody may recognize epitopes within this region and potentially cross-react with isoforms or homologs in other species if conserved.
  • For Western blotting, a band at around 59 kDa is expected; however, post-translational modifications such as glycosylation and GPI-anchor addition may result in a higher apparent molecular weight or multiple bands. Use appropriate reducing conditions and validate in relevant sample systems (e.g., pancreatic lysates or intestinal tissue).
  • For ELISA, the antibody can be used as a capture or detection reagent. Optimize coating concentration and blocking conditions using recombinant GP2 protein or cell lysates.
  • Given the GPI-anchor and membrane association, detergent solubilization may be necessary for efficient extraction.
