Biological Background: GOT2 Function and Localization
- GOT2, also known as mitochondrial aspartate aminotransferase, kynurenine aminotransferase 4 (KAT4), or plasma membrane-associated fatty acid-binding protein (FABPpm), is encoded by the GOT2 gene in humans. It belongs to the class-I pyridoxal-phosphate-dependent aminotransferase family.
- As a key enzyme of the malate-aspartate shuttle, GOT2 is essential for maintaining the intracellular NAD(H) redox balance by facilitating the transfer of reducing equivalents across the mitochondrial membrane.
- GOT2 catalyzes the irreversible transamination of L-kynurenine to kynurenic acid (KA), a neuroactive metabolite, linking it to kynurenine pathway modulation and neurological processes.
- The enzyme also plays a role in cellular long-chain fatty acid uptake through its identity as FABPpm at the plasma membrane, contributing to lipid transport.
- GOT2 is primarily localized to the mitochondrial matrix; however, a fraction is also detected at the cell membrane, consistent with its dual functions in mitochondrial metabolism and fatty acid uptake.
- Post-translational modifications include acetylation, methylation, nitration, and phosphorylation, which may regulate its activity and subcellular distribution. The protein is synthesized with a mitochondrial transit peptide that is cleaved to yield the mature form.
- Mutations in GOT2 are associated with developmental and epileptic encephalopathy 82 (DEE82), underscoring its critical role in brain metabolism.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 30–200 of human GOT2, a region that includes the N-terminal segment of the mature mitochondrial protein after transit peptide cleavage. This design may favor recognition of the endogenous, processed form in mitochondria.
- For Western blot applications, consider enriching mitochondrial fractions to improve signal-to-noise ratio, as GOT2 is predominantly mitochondrial. The predicted molecular weight of the precursor is ~47.5 kDa, while the mature protein migrates near 48 kDa.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), validate detection in tissues or cells with high metabolic activity (e.g., liver, heart, brain) where GOT2 expression is abundant. Optimize permeabilization and antigen retrieval steps to ensure access to mitochondrial epitopes.
- For ELISA, use recombinant GOT2 or tissue lysates as a standard to establish assay sensitivity and specificity. Cross-reactivity with mouse and rat GOT2 has been observed, making it suitable for translational models.
