Biological Background: GOT1 Function and Localization

  • GOT1 is a cytoplasmic aspartate aminotransferase that also exhibits cysteine transaminase activity, commonly referred to as cAST, SGOT, or cCAT. It belongs to the class-I pyridoxal-phosphate-dependent aminotransferase family.
  • It catalyzes the reversible conversion of L-aspartate and α-ketoglutarate to oxaloacetate and L-glutamate, playing a key role in regulating levels of the major excitatory neurotransmitter glutamate in the central nervous system (PubMed:21900944). Related references: PMID:21900944
  • Through its cysteine transaminase activity, GOT1 generates mercaptopyruvate, which is subsequently converted to hydrogen sulfide (H₂S) by 3-mercaptopyruvate sulfurtransferase. H₂S serves as an important synaptic modulator and neuroprotectant in the brain. Related references: PMID:21900944
  • GOT1 additionally catalyzes the formation of (2S)-2-aminobutanoate, a by-product in the cysteine biosynthesis pathway (PubMed:27827456). Related references: PMID:27827456

Experimental Guidance and Technical Tips

  • This antibody was raised against a synthetic peptide immunogen located within amino acids 150–250 of human GOT1. When designing experiments, verify sequence homology if using non-human species, as cross-reactivity with mouse and rat has been observed.
  • For Western blotting, a band at approximately 46 kDa corresponding to GOT1 is expected. Consider optimizing sample preparation and antibody dilution for your specific sample type.
  • In immunofluorescence/immunocytochemistry, GOT1 should predominantly localize to the cytoplasm. Include appropriate fixation and permeabilization steps to ensure antibody accessibility.
  • For ELISA applications, the antibody may be suitable as both capture and detection reagent; however, pairing with a matched antibody is recommended and should be validated in the user’s assay system.
