Biological Background: Aspartate Aminotransferase, Cytoplasmic (GOT1) Function and Localization

  • GOT1 (glutamic-oxaloacetic transaminase 1, cytoplasmic) is also known as AST, AST1, SGOT, cCAT, cAspAT, and GIG18. It is a pyridoxal-phosphate-dependent aminotransferase.
  • Its primary reaction is the reversible interconversion of L-aspartate and 2-oxoglutarate to oxaloacetate and L-glutamate, linking amino acid and carbohydrate metabolism via the malate-aspartate shuttle.
  • In the brain, GOT1 acts as a glutamate scavenger, helping regulate excitatory neurotransmission and contributing to neuroprotection. Related references: PMID:21900944
  • Using L-cysteine as a substrate, GOT1 produces mercaptopyruvate, a key precursor for hydrogen sulfide (H2S) synthesis. H2S acts as a synaptic modulator and neuroprotectant. Related references: PMID:21900944 PMID:27827456

Experimental Guidance and Technical Tips

  • The immunogen used to generate this polyclonal antibody corresponds to amino acids 244–413 of human GOT1, a region outside the active site; thus, the antibody may recognize both native and denatured forms of the protein.
  • For Western blot (WB), a band at approximately 46 kDa is expected. Consider using whole-cell lysates from tissues rich in GOT1, such as liver or brain, as positive controls.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), anticipate cytoplasmic staining. Optimize antigen retrieval and permeabilization protocols as needed.
  • When performing ELISA, titrate the antibody to determine optimal capture/detection conditions. This antibody may be used in combination with matching antigen or detection antibodies for sandwich ELISA format.
  • Given cross-reactivity with mouse and rat, this antibody is suitable for translational studies in these model organisms; validate in the relevant sample system to confirm specificity.
