Biological Background: GOLGA1 Function and Localization

  • GOLGA1, also known as golgin-97, is a member of the golgin family of coiled-coil proteins associated with the Golgi apparatus.
  • It plays a key role in vesicular trafficking at the Golgi, particularly in endosome-to-Golgi transport by tethering TBC1D23-bound vesicles to the trans-Golgi network. Related references: PMID:29084197, PMID:38552021
  • Selectively regulates E-cadherin transport from the trans-Golgi network in tubulovesicular carriers, influencing cell-cell adhesion. Related references: PMID:34969853
  • Subcellular localization: primarily found at the Golgi apparatus membrane, trans-Golgi network membrane, and also in cytoplasmic vesicles like secretory vesicles and acrosomes.
  • Contains coiled-coil domains, and is subject to ADP-ribosylation and phosphorylation (UniProt keywords).
  • (Microbial infection) Plays a role in poxvirus morphogenesis by translocating to viral factories to facilitate membrane transport for virion assembly.
  • The protein has a predicted molecular weight of ~88 kDa (UniProt: 88184 Da).

Experimental Guidance and Technical Tips

  • Designed using a recombinant immunogen spanning the N-terminal region (aa 1-300), which is expected to recognize endogenous GOLGA1 in multiple species due to high cross-reactivity with human, mouse, and rat.
  • For Western blotting (WB), a band around 88 kDa is expected. Consider optimizing antibody dilution for your specific sample type.
  • For immunofluorescence/immunocytochemistry (IF/ICC), GOLGA1 localizes to the Golgi apparatus; a perinuclear, Golgi-like staining pattern should be observed. Co-staining with a Golgi marker is suggested.
  • ELISA application can be used for antibody titration or antigen detection; validate in the relevant sample system.
  • Store at recommended conditions (shipped on ice bag, likely -20°C for long-term storage). Avoid repeated freeze-thaw cycles.
