Biological Background: Alpha-1,6-mannosylglycoprotein 6-beta-N-acetylglucosaminyltransferase A Function and Localization
- MGAT5 (GNT-V) is an alpha-1,6-mannosylglycoprotein 6-beta-N-acetylglucosaminyltransferase A that belongs to the glycosyltransferase family.
- Catalyzes the addition of N-acetylglucosamine in beta1-6 linkage to alpha-linked mannose of biantennary N-linked oligosaccharides. Related references: PMID:10395745, PMID:30140003
- Essential for the biosynthesis of branched complex-type N-glycans on receptors such as EGFR, TGFR, and CDH2, thereby modulating growth factor signaling. Related references: PMID:10395745, PMID:22614033, PMID:30140003
- Regulates cell adhesion and migration by modifying CDH2 glycosylation, which inhibits homotypic cell-cell adhesion.
- Plays a role in inflammation: MGAT5-dependent TCR glycosylation limits TCR clustering and dampens antigen responses, while it also inhibits monocyte adhesion and diapedesis. Related references: PMID:22614033
- Promotes proliferation of endothelial cells and angiogenesis by facilitating FGF2 release from extracellular matrix.
- Subcellular localization: Golgi apparatus membrane and secreted.
- Post-translational modifications: glycoprotein, contains disulfide bonds.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein corresponding to amino acids 627-741 of human GNT-V/MGAT5 (NP_002401.1). Consider this region's conservation when expecting cross-reactivity in mouse.
- This polyclonal antibody is suitable for Western blot (WB) and ELISA applications. For WB, an expected band around 85 kDa is observed in human samples; validate in mouse lysates due to cross-reactivity.
- Due to the polyclonal nature, the antibody may detect multiple isoforms or degradation products; optimize the antibody dilution for your specific experimental conditions.
- Pre-validate the antibody in the relevant sample system (e.g., cell lines, tissues) to confirm specificity, especially if studying glycosylation-dependent events.
