Biological Background: GNPDA1 Function and Localization
- GNPDA1 (glucosamine-6-phosphate deaminase 1) is also known as glucosamine-6-phosphate isomerase 1, oscillin, GNPI, HLN, and KIAA0060. It is encoded by the GNPDA1 gene in humans (Gene ID: 10007).
- Catalyzes the reversible deamination and aldo-keto isomerization of alpha-D-glucosamine 6-phosphate to beta-D-fructose 6-phosphate and ammonium, a key regulatory step in the hexosamine pathway for UDP-GlcNAc biosynthesis. Related references: PMID:21807125 PMID:26887390
- At high ammonium concentrations, drives the amination and isomerization of fructose 6-phosphate toward hexosamines, influencing UDP-GlcNAc synthesis. Related references: PMID:21807125 PMID:26887390
- Plays a role in fine-tuning metabolic fluctuations of cytosolic UDP-GlcNAc and their effects on hyaluronan synthesis during tissue remodeling. Related references: PMID:26887390
- Might trigger calcium oscillations in mammalian eggs, essential for egg activation and early embryonic development (By similarity).
- Localizes to the cytoplasm.
- Belongs to the glucosamine-6-phosphate deaminase family; post-translational modifications include acetylation and phosphorylation as indicated by proteomics.
- Alternative splicing results in multiple transcript variants.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein covering amino acids 1-70 of human GNPDA1 (N-terminal region). This N-terminal targeting may contribute to the antibody's specificity.
- This antibody is validated for Western blot (WB) and ELISA. For WB, consider using a protein weight of approximately 33 kDa as a reference for GNPDA1.
- Cross-reactivity is reported for human, mouse, and rat; thus, it can be considered for studies in these species, but validation in the specific sample system is recommended.
- For functional studies of the hexosamine pathway or hyaluronan synthesis, this antibody may aid in detecting GNPDA1 expression.
