Biological Background: GNAQ Function and Localization

  • GNAQ encodes the guanine nucleotide-binding protein G(q) subunit alpha, a transducer in GPCR signaling cascades. Related references: PMID:34556863, PMID:35672283, PMID:37991948
  • The protein alternates between inactive (GDP-bound) and active (GTP-bound) states; GPCR activation promotes GDP release and GTP binding, while intrinsic GTPase activity returns it to the inactive state. Related references: PMID:37991948
  • Activated GNAQ stimulates phospholipase C-beta (PLCB1-4), leading to production of second messengers diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3). Related references: PMID:37991948
  • GNAQ is required for platelet activation, B-cell selection and survival, and chemotaxis of neutrophils and dendritic cells (by similarity).
  • GNAQ transduces FFAR4 signaling in response to long-chain fatty acids. Related references: PMID:27852822
  • Subcellular localization includes the cell membrane, Golgi apparatus, nucleus, and nuclear membrane.
  • Predominantly expressed in ovary, prostate, testis, and colon; down-regulated in peripheral blood lymphocytes of rheumatoid arthritis patients (at protein level).
  • Together with GNA11, required for heart development (by similarity).

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 50–150 of human GNAQ, a conserved region among human, mouse, and rat; consider verifying cross-reactivity in your target species and application.
  • For Western blotting, consider using whole-cell lysates from tissues with high GNAQ expression (ovary, prostate, testis, colon) and validate with a loading control.
  • For IHC-P, antigen retrieval may be necessary given the nuclear and multiple membrane localizations; check staining in positive and negative tissue controls.
  • ELISA performance should be validated using recombinant protein or peptide to establish optimal coating and detection conditions.
