Biological Background: GNAI2 Function and Localization

  • GNAI2 encodes the guanine nucleotide-binding protein G(i) subunit alpha-2 (adenylate cyclase-inhibiting G alpha protein), a member of the G protein alpha subunit family that functions as a molecular switch in signal transduction.

  • Inhibits adenylate cyclase activity of ADCY1, ADCY5, and ADCY6, leading to decreased intracellular cAMP, and is involved in hormonal regulation of adenylate cyclase in response to beta-adrenergic stimuli.

  • The immunogen is a full-length recombinant human GNAI2 protein (amino acids 1-355), which likely generates antibodies targeting multiple epitopes across the entire protein. When designing assays, consider that the antibody may recognize both denatured and native conformations, as the immunogen is a recombinant fusion protein.
  • For Western blotting, validate in human and mouse cell lines or tissues known to express GNAI2, such as endothelial cells. A specific band at approximately 40 kDa should be detected; however, signal intensity may vary depending on sample preparation and expression levels.
  • In ELISA applications, use recombinant GNAI2 protein or cell lysates as coating antigens; perform checkerboard titrations to determine optimal antibody and antigen concentrations.
