Biological Background: GNA15 Function and Localization
- Guanine nucleotide-binding protein subunit alpha-15 (GNA15), also known as G alpha-16 or G protein subunit alpha-16, is a member of the G protein alpha subunit family that functions as a molecular switch in intracellular signaling.
- Transduces signals from G protein-coupled receptors (GPCRs) to downstream effectors, playing a crucial role in various cellular responses.
- Collaborates with the adapter protein TTC1 to promote HRAS activation and ERK1/2 phosphorylation independently of phospholipase Cbeta signaling. Related references: PMID:12748287, PMID:20639119
- Initiates EMR2-mediated signaling, leading to activation of Akt, MAPK, and NF-kappa-B, which drives macrophage differentiation and inflammatory responses. Related references: PMID:28421075
- Specifically expressed in hematopoietic cells, with protein expression detected in epididymis.
- Characterized by ADP-ribosylation, GTP-binding, magnesium and metal binding, nucleotide-binding, and transducer activities, as annotated by UniProtKB keywords.
- Encoded by the GNA15 gene (also designated GNA16), with a predicted molecular weight of approximately 43.5 kDa and an observed weight of 44kDa.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning the full-length human GNA15 (amino acids 1-374), which may allow recognition of multiple epitopes.
- For Western blot (WB), the predicted molecular weight is ~44kDa; validate band size in your specific sample type, preferably using hematopoietic cell lines due to tissue specificity.
- In ELISA applications, consider optimizing coating antigen concentration and detection conditions to achieve specific signal.
- Cross-reactivity with mouse and rat suggests potential utility in rodent model studies; however, perform species-specific validation.
- As a polyclonal antibody, monitor lot-to-lot consistency when used in quantitative or longitudinal experiments.
