Biological Background: GM130 Function and Localization

  • GM130 (Golgin subfamily A member 2) is a peripheral membrane protein of the cis-Golgi stack, acting as a membrane skeleton that maintains Golgi structure and facilitates vesicle tethering (Probable) (PubMed:16489344). Related references: PMID:16489344
  • Together with p115/USO1 and STX5, it mediates vesicle tethering and fusion at the cis-Golgi membrane to maintain the stacked and interconnected structure of the Golgi apparatus.
  • GM130 is essential for mitotic Golgi disassembly: phosphorylation at Ser-37 by CDK1 at mitosis onset inhibits p115 interaction, preventing COPI vesicle tethering and halting transport.
  • It plays a key role in spindle pole assembly and centrosome organization (PubMed:26165940). GM130 promotes spindle pole assembly by recruiting TPX2 to Golgi membranes, which activates AURKA kinase and stimulates local microtubule nucleation, thereby linking Golgi membranes to the mitotic spindle (PubMed:19242490, PubMed:26165940). Related references: PMID:26165940 PMID:19242490
  • It also regulates centrosome organization independently of its role in Golgi structure (PubMed:18045989, PubMed:19109421). Related references: PMID:18045989 PMID:19109421
  • GM130 is required for Golgi ribbon formation and proper glycosylation of membrane and secretory proteins (PubMed:16489344, PubMed:17314401). Related references: PMID:16489344 PMID:17314401
  • Subcellularly, GM130 localizes to the cis-Golgi network membrane, ER-Golgi intermediate compartment membrane, and the spindle pole region of the cytoskeleton.
  • GM130 features coiled-coil domains and undergoes phosphorylation and methylation; mutations in GOLGA2 are associated with intellectual disability.

Experimental Guidance and Technical Tips

  • The immunogen (aa 20–300 of human GM130) encompasses the N-terminal coiled-coil region involved in p115 binding; consider this when interpreting band patterns or potential isoform recognition in Western blot.
  • For Western blot, the predicted molecular weight is ~113 kDa; use appropriate gel percentage (e.g., 6–8% SDS-PAGE) and efficient transfer protocols for high-molecular-weight proteins.
  • In IHC-P and IF/ICC applications, optimize antigen retrieval and permeabilization steps to ensure accessibility to the cis-Golgi-localized target; validate in your specific tissue or cell system.
  • When using ELISA, assess the antibody’s suitability as capture or detection reagent; cross-reactivity with mouse and rat GM130 may be advantageous for multispecies studies.
  • As a polyclonal antibody, lot-to-lot consistency should be verified for critical quantitative assays, and appropriate controls are recommended to confirm specificity.
