Biological Background: Sodium- and chloride-dependent glycine transporter 2 Function and Localization
- Sodium- and chloride-dependent glycine transporter 2 (Glyt2), also known as Solute carrier family 6 member 5, is a transmembrane protein encoded by the SLC6A5 gene.
- Mediates high-affinity sodium- and chloride-dependent reuptake of glycine into presynaptic terminals, terminating glycinergic neurotransmission at strychnine-sensitive synapses. Related references: PMID:10381548, PMID:10606742, PMID:16751771, PMID:9845349
- Subcellularly localized to the cell membrane, consistent with its role in neurotransmitter transport.
- Predominantly expressed in the medulla, with lower levels detected in the spinal cord and cerebellum.
- Exhibits features including alternative splicing, N-linked glycosylation, and phosphorylation; it is a disulfide-bonded membrane glycoprotein involved in sodium/chloride symport.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 250–350 of human Glyt2 (Q9Y345); ensure target region is conserved in your experimental species.
- For Western blotting, use appropriate protein loading controls around 87 kDa and consider pre-absorption controls to confirm specificity.
- In ELISA, titrate both the primary antibody and antigen to optimize signal-to-noise ratio; include appropriate blank and negative controls.
- Because Glyt2 expression is tissue-specific, validate sensitivity and specificity in relevant samples (e.g., medulla or spinal cord lysates) before large-scale experiments.
