Biological Background: Glutamine Synthetase Function and Localization
- Glutamine synthetase (GLUL), also known as glutamate-ammonia ligase, GS, GLNS, PIG43, and PIG59, is a key enzyme encoded by the GLUL gene.
- Catalyzes the ATP-dependent conversion of glutamate and ammonia to glutamine, a critical reaction for nitrogen metabolism and neurotransmitter regulation (PubMed:16267323, PubMed:30158707, PubMed:36289327). Related references: PMID:16267323 PMID:30158707 PMID:36289327
- In the brain, it converts neurotoxic glutamate to harmless glutamine and regulates ammonia levels; in the liver, it participates in ammonia removal (By similarity).
- Functions in ammonium detoxification during erythropoiesis, removing ammonium generated by porphobilinogen deaminase during heme biosynthesis (By similarity).
- Essential for proliferation of fetal skin fibroblasts (PubMed:18662667). Related references: PMID:18662667
- Independently of its glutamine synthetase activity, acts as a palmitoyltransferase for RHOJ, regulating endothelial cell migration during vascular development (PubMed:30158707). Related references: PMID:30158707
- Plays a role in ribosomal 40S subunit biogenesis (PubMed:26711351). Related references: PMID:26711351
- Interacts with BEST2, inhibiting its channel activity in the absence of intracellular L-glutamate but sensitizing it to L-glutamate, thereby regulating ion channel gating (PubMed:36289327). Related references: PMID:36289327
- Localizes to the cytoplasm, mitochondrion, and cell membrane, and is expressed in endothelial cells.
Experimental Guidance and Technical Tips
- For Western blotting, a band corresponding to the predicted molecular weight of approximately 42 kDa should be detected; verify with appropriate positive controls.
- The immunogen is a synthetic peptide from a conserved region (aa 200-300), suggesting potential cross-reactivity across human, mouse, and rat; validate in your specific sample system.
- This antibody is recommended for WB, IHC-P, and ELISA; optimize antibody dilution and incubation conditions for your assay format.
- Consider using protein extraction protocols that preserve post-translational modifications (acetylation, palmitoylation) if studying these aspects.
