- GLUT4, encoded by the SLC2A4 gene, is a member of the solute carrier family 2 (facilitated glucose transporter) and is also known as glucose transporter type 4.
- It functions as an insulin-regulated facilitative glucose transporter, critical for removing glucose from the bloodstream.
- In the absence of insulin, GLUT4 is efficiently retained in intracellular storage compartments within muscle and fat cells.
- Upon insulin stimulation, GLUT4 translocates from these compartments to the cell surface, enabling glucose influx.
- Subcellular localization includes the cell membrane, endomembrane system, cytoplasm, and perinuclear region.
- Tissue specificity: predominantly expressed in skeletal and cardiac muscles, as well as brown and white adipose tissue.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from residues 210–310; therefore, the antibody is expected to recognize linear epitopes, suitable for denaturing Western blot protocols.
- For WB, use standard denaturing and reducing conditions; expect a specific band at approximately 55 kDa (theoretical MW ~54.8 kDa) in human and mouse samples.
- For IF/ICC and IHC-P, consider testing both untreated and insulin-stimulated cells/tissues to observe GLUT4 translocation.
