Biological Background: GLUT4 Function and Localization

  • GLUT4, encoded by the SLC2A4 gene, is a member of the solute carrier family 2 (facilitated glucose transporter) and is also known as glucose transporter type 4, insulin-responsive (GLUT-4).
  • It functions as an insulin-regulated facilitative glucose transporter, crucial for clearing glucose from the bloodstream. In the absence of insulin, GLUT4 is retained intracellularly in storage compartments of muscle and fat cells; upon insulin stimulation, it translocates to the cell surface to transport glucose.
  • Subcellular localization includes the cell membrane, endomembrane system, and cytoplasm, particularly in the perinuclear region.
  • Tissue expression is predominantly in skeletal and cardiac muscles, and brown and white adipose tissue.
  • Post-translational modifications include palmitoylation, phosphorylation, and ubiquitin conjugation.
  • The protein is associated with glucose transport and diabetes mellitus, and alternative splicing generates isoforms.
  • Three-dimensional structural data are available for this transporter.
  • It is a multi-pass transmembrane glycoprotein involved in proteomics identification and classified as a reference proteome.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a C-terminal intracellular region (amino acids 410–509), which may assist in detecting both intracellular and membrane-localized pools depending on permeabilization.
  • For Western blot, bands at ~55 kDa (glycosylated) and ~44 kDa (non-glycosylated) may be observed; consider using glycosidase treatment to confirm specificity.
  • In IF-P and IHC-P, validate with appropriate controls and consider insulin stimulation to visualize translocation from intracellular compartments to the plasma membrane.
  • For ELISA, ensure antibody pair compatibility if used in sandwich assays; test across insulin-treated vs. untreated cell lysates to assess performance.
