Biological Background: GLUT2/SLC2A2 Function and Localization
- Solute carrier family 2, facilitated glucose transporter member 2 (GLUT2/SLC2A2) is a member of the GLUT family of facilitative sugar transporters.
- It mediates the transport of hexoses including glucose, fructose, and galactose across the cell membrane (PubMed:16186102, PubMed:23396969, PubMed:28083649). Related references: PMID:16186102 PMID:23396969 PMID:28083649
- In hepatocytes and pancreatic beta cells, GLUT2 facilitates bidirectional glucose transport, contributing to glucose sensing and homeostasis (PubMed:8027028). Related references: PMID:8027028
- It may also participate in transcellular glucose transport in the small intestine and kidney, cooperating with Na+/glucose cotransporters (PubMed:3399500). Related references: PMID:3399500
- GLUT2 has been shown to transport dehydroascorbate and urate (PubMed:23396969, PubMed:40209957). Related references: PMID:23396969 PMID:40209957
- The protein is localized to the cell membrane and is primarily expressed in liver, insulin-producing beta cells, small intestine, and kidney.
- Post-translational modifications include glycosylation and phosphorylation; the SLC2A2 gene undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the N-terminal region (amino acids 1–100) of human GLUT2/SLC2A2; consider that this may enable detection of multiple isoforms if the epitope is conserved.
- For Western blot (WB), expect a band around 57 kDa; validate expression in relevant cell lines or tissues such as liver or pancreatic beta cells.
- For immunofluorescence (IF) and immunohistochemistry (IHC-P), appropriate antigen retrieval and permeabilization methods should be optimized; consider using positive control tissues like liver or pancreas.
- For ELISA, the antibody may be used as a capture or detection reagent; titrate and validate against recombinant or endogenous targets in the relevant sample matrix.
