Biological Background: GluR1/GRIA1 Function and Localization
- Glutamate receptor 1 (GluR1/GRIA1) is an AMPA-selective ionotropic glutamate receptor, also known as GluR-A, GluR-K1, or AMPA-selective glutamate receptor 1, encoded by the GRIA1 gene. It belongs to the glutamate-gated ion channel family and mediates fast excitatory neurotransmission in the central nervous system.
- Functions as a ligand-gated ion channel activated by L-glutamate and the synthetic agonist AMPA, as well as quisqualic acid and kainic acid. Upon agonist binding, the channel opens, allowing influx of monovalent (Na⁺, K⁺, Li⁺) and divalent (Ca²⁺, Mg²⁺, Sr²⁺) cations, thereby depolarizing the postsynaptic membrane (PubMed:1311100, PubMed:20805473). - The immunogen is a synthetic peptide corresponding to amino acids 800–906 within the intracellular C‑terminal domain of human GluR1/GRIA1. This region is conserved across mouse and rat, supporting the observed cross‑reactivity.
- For Western blot, expect a band at approximately 102 kDa. Due to glycosylation and other modifications, slight shifts may occur; validate using appropriate positive and negative controls.
- For ELISA, the antibody may be used as a capture or detection reagent; consider pairing with a second antibody recognizing a distinct epitope or using recombinant GluR1 protein as a standard.
- Since the antibody recognizes a linear epitope, denaturing and reducing conditions in WB should be effective. However, for native detection (e.g., immunohistochemistry), test under non‑denaturing conditions and include appropriate tissue controls.
