Biological Background: GLUD1/2 Function and Localization
- GLUD1 (glutamate dehydrogenase 1) is a mitochondrial matrix enzyme that catalyzes the reversible oxidative deamination of L-glutamate to alpha-ketoglutarate and ammonia, using NAD(P)+ as a cofactor.
- It plays a key role in glutamine anaplerosis, producing alpha-ketoglutarate to replenish tricarboxylic acid cycle intermediates. Related references: PMID:11032875 PMID:11254391 PMID:16023112
- The enzyme is involved in insulin homeostasis, regulating glucose-stimulated insulin secretion in pancreatic beta cells. Related references: PMID:11297618 PMID:9571255
- It may modulate the turnover of the excitatory neurotransmitter glutamate, potentially influencing learning and memory processes.
- GLUD1 localizes primarily to the mitochondrial matrix and has also been detected at the endoplasmic reticulum.
- The protein undergoes post-translational modifications including acetylation, ADP-ribosylation, and phosphorylation, which regulate its activity.
- This antibody targets both GLUD1 and its isoform GLUD2 (also known as GDH2), due to the high sequence homology between the two proteins; GLUD2 is predominantly expressed in neural and testicular tissues.
Experimental Guidance and Technical Tips
- This rabbit monoclonal antibody was raised against a recombinant protein covering amino acids 319–558 of human GLUD1/2, a region conserved across both isoforms. It is expected to detect both endogenous GLUD1 and GLUD2.
- For Western blotting, a band at approximately 61 kDa should be observed in lysates from human, mouse, and rat samples. Use appropriate positive controls such as liver or brain tissue extracts.
- In immunostaining (IF/ICC, IHC-P), mitochondrial localization is anticipated. A punctate perinuclear pattern may be visible; counterstaining with a mitochondrial marker can help confirm specificity.
- The antibody is applicable to WB, IF/ICC, IF-P, IHC-P, and ELISA. Optimal working concentrations and conditions should be determined empirically for each assay and sample type.
- Due to the high similarity between GLUD1 and GLUD2, cross-reactivity with both targets is expected; this should be considered when interpreting results in tissues where both isoforms are expressed.
