Biological Background: Glucosylceramidase beta (GBA) Function and Localization

  • Catalyzes lysosomal hydrolysis of glucosylceramides (GlcCers) into free ceramide and glucose, a critical step in sphingolipid catabolism. Related references: PMID:15916907, PMID:24211208, PMID:32144204
  • Essential for the degradation of complex lipids and turnover of cellular membranes. Related references: PMID:27378698
  • Participates in the PKC-activated salvage pathway of ceramide formation by providing ceramide from GlcCer breakdown. Related references: PMID:19279011
  • Catalyzes transglucosylation of cholesterol, using GlcCer as glucose donor, to form cholesteryl glucoside, and can also hydrolyze cholesteryl glucoside back to cholesterol and glucose. Related references: PMID:24211208, PMID:26724485, PMID:32144204
  • Exhibits alternative glycosidase/transferase activities toward galactosylceramide (GalCer) and xylosylceramide (XylCer) substrates, although with lower activity than with GlcCer. Related references: PMID:32144204, PMID:33361282, PMID:39395789
  • Localized to the lysosome membrane; mutations in GBA lead to Gaucher disease and are associated with Parkinsonism.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 437–536, a region likely to be accessible in both native and denatured GBA.
  • For Western blot, expect a band near 60 kDa; validate specificity using relevant human or rat lysates.
  • For IHC-P, optimize antigen retrieval to unmask the epitope and confirm staining in positive control tissues.
  • For ELISA, consider pairing with a detection antibody and validate using recombinant GBA or known positive samples.
  • Cross-reactivity with rat enables interspecies studies; validate expression levels in the target rat tissues.
