Biological Background: Glucose-6-Phosphate Isomerase Function and Localization

  • Glucose-6-phosphate isomerase (GPI) is also known as autocrine motility factor (AMF), neuroleukin (NLK), phosphoglucose isomerase (PGI), phosphohexose isomerase (PHI), and sperm antigen 36 (SA-36).
  • The enzyme catalyzes the conversion of alpha-D-glucose-6-phosphate to beta-D-fructose-6-phosphate, the second step in glycolysis, and the reverse reaction in gluconeogenesis, within the cytoplasm. Related references: PMID:28803808
  • GPI also exhibits C2-epimerase activity, interconverting D-glucose-6-phosphate and D-mannose-6-phosphate, and anomerase activity for multiple hexose-6-phosphates.
  • Besides its metabolic role, extracellular GPI functions as a cytokine: as autocrine motility factor (AMF), a secreted angiogenic factor enhancing endothelial cell motility. Related references: PMID:11437381
  • It also acts as neuroleukin, a neurotrophic factor supporting spinal and sensory neuron survival, and is released by lectin-stimulated T-cells to induce immunoglobulin secretion. Related references: PMID:11004567 PMID:3352745
  • Subcellularly, GPI is found in the cytoplasm and is secreted.
  • Post-translational modifications include acetylation, hydroxylation, phosphorylation, and ubiquitin conjugation; alternative splicing produces multiple isoforms.
  • Mutations in GPI are associated with hereditary hemolytic anemia.

Experimental Guidance and Technical Tips

  • The antibody was generated using a recombinant immunogen encompassing amino acids 1-280 of human GPI (NP_000166.2), which may target conserved epitopes; cross-reactivity with mouse and rat is confirmed.
  • For Western blotting (WB), a band around 63 kDa is expected; consider using reducing conditions and appropriate positive controls such as human, mouse, or rat cell lysates.
  • In immunofluorescence/immunocytochemistry (IF/ICC), note that GPI is cytoplasmic and secreted; fixation and permeabilization may be required for intracellular detection, and consider co-staining with organelle markers.
  • For ELISA, the antibody may be used as capture or detection reagent; validate with recombinant GPI protein or biological samples to establish linearity and sensitivity.
  • As a monoclonal antibody, batch-to-batch consistency is expected, but always optimize working dilutions for your specific assay and sample type.
  • The antibody is labeled as KD validated; consider incorporating appropriate knockdown controls in your experiments to confirm specificity.
