Biological Background: Glucocorticoid Receptor Function and Localization
- The glucocorticoid receptor (GR), encoded by NR3C1, is a member of the nuclear receptor family. It exists as multiple isoforms generated by alternative splicing and alternative translation initiation, including the canonical alpha isoform and the beta isoform that acts as a dominant negative inhibitor. Related references: PMID:20484466, PMID:7769088, PMID:8621628
- GR functions as a ligand-dependent transcription factor that binds to glucocorticoid response elements (GRE) to regulate gene expression, playing critical roles in cellular proliferation, differentiation, and inflammatory responses. Related references: PMID:27120390, PMID:37478846, PMID:28139699
- The receptor contributes to chromatin remodeling and facilitates accurate chromosome segregation during mitosis, potentially acting as a tumor suppressor. Related references: PMID:9590696, PMID:25847991
- GR can mediate rapid mRNA degradation by binding to the 5' UTR of target mRNAs and recruiting the RNA helicase UPF1 and decapping enzyme DCP1A. Related references: PMID:25775514
- It mediates glucocorticoid-induced apoptosis and exhibits both transcriptional activation and repression activities. Related references: PMID:23303127, PMID:11435610, PMID:15769988, PMID:15866175
- Subcellularly, GR is primarily cytoplasmic but translocates to the nucleus upon ligand binding. It also localizes to mitochondria, the cytoskeleton (spindle, centrosome), and chromosomes.
- Widely expressed in tissues including bone, stomach, lung, liver, colon, breast, ovary, pancreas, kidney, and various heart regions. Related references: PMID:25847991, PMID:10902803
- Post-translational modifications include acetylation, phosphorylation, methylation, and ubiquitination; it is also annotated with keywords like DNA-binding, zinc-finger, metal-binding, and steroid-binding.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-280 of human Glucocorticoid Receptor (NP_001191194.1). This N-terminal region may be suitable for detecting multiple isoforms.
- For Western blot, a band around 86 kDa is expected; validate in relevant cell or tissue lysates.
- IHC-P and IF/ICC applications may require antigen retrieval optimization; include appropriate positive and negative controls.
- Given the wide tissue expression, antibody specificity should be confirmed using knockout or knockdown models if possible.
- Cross-reactivity with mouse and rat GR enables multiorganism studies; consider species-specific sequence alignment for binding confidence.
