Biological Background: Glutaredoxin-3 Function and Localization

  • Glutaredoxin-3 (GLRX3), also known as PICOT, TXNL2, GRX3, GRX4, GLRX4, and TXNL3, is a member of the glutaredoxin family; however, it lacks thioredoxin activity due to the absence of a conserved catalytic motif.
  • Together with BOLA2, GLRX3 serves as a cytosolic iron-sulfur (Fe-S) cluster assembly factor, facilitating [2Fe-2S] cluster insertion into a subset of cytosolic proteins. Related references: PMID:26613676 PMID:27519415
  • It acts as a critical negative regulator of cardiac hypertrophy and a positive inotropic regulator (by similarity).
  • GLRX3 is required for hemoglobin maturation. Related references: PMID:23615448
  • Subcellularly, GLRX3 is located in the cytoplasm, including the cytosol, cell cortex, and myofibril sarcomere Z line.
  • It is highly expressed in heart, spleen, and testis, with lower expression in thymus, peripheral blood leukocytes, and weak expression in lung, placenta, colon, and small intestine.

  • The immunogen is a recombinant fusion protein spanning the full-length human GLRX3 (amino acids 1-335). For Western blot, the predicted molecular weight is approximately 37 kDa; consider verifying band size in your sample system.
  • For immunoprecipitation, this polyclonal antibody may enrich endogenous GLRX3; validate the IP efficiency with a suitable lysis buffer and detection antibody.
  • ELISA application is possible; consider testing serial dilutions of the antibody against the immunogen or target protein to establish optimal working conditions.
  • Since cross-reactivity is reported for human, validate reactivity in other species if required, as the antibody may not cross-react with mouse or rat GLRX3.
