Biological Background: Glomulin Function and Localization

  • Glomulin (GLMN) is a regulatory component of cullin-RING-based SCF E3 ubiquitin-protein ligase complexes. It inhibits E3 ligase activity by binding to RBX1 and disrupting its interaction with the E2 enzyme CDC34, thereby inhibiting RBX1-mediated neddylation of CUL1.   • It is required for normal stability and cellular levels of key SCF complex components, including FBXW7, RBX1, CUL1, CUL2, CUL3, and CUL4A, contributing to the regulation of CCNE1 and MYC levels.

  • Contributes to the regulation of RPS6KB1 phosphorylation. Related references: PMID:11571281
  • Ubiquitously expressed in human tissues.
  • Post-translationally modified by acetylation and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 405–594) of human Glomulin. This region may confer specificity for isoforms containing this sequence; consult sequence alignments for cross-reactivity predictions.
  • For Western blotting, a band at approximately 68 kDa is expected. Optimize loading controls and antibody dilution using positive control lysates from human or mouse tissues/cells known to express GLMN.
  • For ELISA, consider coating with the recombinant immunogen or native protein in the appropriate buffer system, and validate with known standards.
  • Since the antibody is polyclonal, batch-to-batch consistency should be monitored; it is recommended to pre-adsorb or affinity-purify if needed for highly specific assays.
