Biological Background: GLI3 Function and Localization
- GLI3 is a transcription factor acting as a key effector of the smoothened signaling pathway, crucial for the development and patterning of body structures including the brain and limbs. Related references: PMID:10075717 PMID:10375510 PMID:10693759
- It binds specifically to the minimal GLI-consensus sequence 5'-GACCACCCA-3' and functions both as a transcriptional activator (full-length GLI3) and repressor (GLI3R).
- The transcription repressor form GLI3R is the major form in the absence of hedgehog signaling, generated by proteolytic processing of full-length GLI3. Related references: PMID:10375510 PMID:10693759
- The full-length GLI3 activator form is a minor form induced by smoothened activation, and cooperates with TRPS1 to regulate chondrocyte proliferation and PTHLH expression. Related references: PMID:10075717 PMID:27146893 PMID:19389374
- Localizes to the nucleus, cytoplasm, and cell projections including the primary cilium, consistent with its role in signal transduction.

Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against an internal peptide (aa 681-788) and may recognize both full-length GLI3 (activator) and the proteolytically processed repressor form (GLI3R). For Western blot, an expected band around 170 kDa corresponds to the full-length protein; smaller fragments may also be visible.
- For immunofluorescence/immunocytochemistry and immunohistochemistry, consider including appropriate controls (e.g., siRNA knockdown or known positive/negative tissues) to confirm specificity. Subcellular patterns should include nuclear, cytoplasmic, and ciliary staining.
- ELISA applications can be developed using the antibody as capture or detection reagent, but optimal dilutions and pairings should be validated in the relevant sample system.
