Biological Background: GLI2 Function and Localization
- GLI2 is a zinc-finger transcription factor of the GLI family that serves as a key effector of the hedgehog signaling pathway, playing a critical role in skeleton development. Related references: PMID:15994174 PMID:18455992 PMID:19878745
- As a transcriptional activator, GLI2 binds to the GLI-consensus sequence 5'-GACCACCCA-3' and activates target gene expression upon smoothened (SMO) stimulation. Related references: PMID:18455992 PMID:19878745 PMID:24311597
- A proteolytically processed repressor form (GLI2R) is generated in the absence of hedgehog signal, constituting a minor pool and inhibiting target gene transcription (By similarity).
- GLI2 localizes to the nucleus, cytoplasm, and cell projections including the cilium, which is essential for hedgehog signal transduction.
- GLI2 undergoes multiple post-translational modifications, including acetylation, phosphorylation, and ubiquitination, as indicated by keyword annotations.
- In HTLV-1-infected T-cells, GLI2 acts as a Tax-dependent transcriptional activator by binding the Tax-responsive element (TRE-2S) to augment viral enhancer activity. Related references: PMID:9557682
- GLI2 is expressed in breast cancers and in HTLV-1-infected T-cell lines, with isoform-specific expression patterns. Related references: PMID:26565916 PMID:9557682
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-175) of mouse GLI2; it may detect both activator and repressor forms. Consider verifying specific band patterns in your model system.
- For Western blot, optimize antibody dilution using lysates from mouse or rat tissues known to express GLI2.
- For ELISA, validate assay conditions with recombinant GLI2 protein or peptide antigens to ensure specific detection.
- Cross-reactivity is reported for mouse and rat; confirm species specificity if using other models.
