Biological Background: Transcription activator GLI2 Function and Localization
- Transcription factor GLI2 is a key effector of the Smoothened (SMO) signaling pathway, essential for skeletal development (PubMed:15994174, PubMed:18455992, PubMed:19878745). Related references: PMID:15994174 PMID:18455992 PMID:19878745
- It binds to the minimal GLI-consensus sequence 5’-GACCACCCA-3’ to regulate target genes, functioning as both a transcriptional activator (full-length GLI2) and repressor (GLI2R). The activator form predominates, while the repressor is generated by proteolytic processing in the absence of SMO signaling.
- GLI2 plays a critical role in Hedgehog (SHH) signaling, activating target gene expression upon SMO stimulation (PubMed:18455992, PubMed:19878745, PubMed:24311597). Related references: PMID:18455992 PMID:19878745 PMID:24311597
- In HTLV-1-infected T-cells, GLI2 acts as a Tax-dependent transcriptional activator, binding to the viral TRE-2S element (5’-GAACCACCCA-3’) to augment enhancer activity (PubMed:9557682). Related references: PMID:9557682
- Subcellular localization includes the nucleus, cytoplasm, and cilia. It is expressed in breast cancers and HTLV-1-infected T-cell lines, with multiple isoforms showing tissue-specific expression.
- Post-translational modifications encompass acetylation, phosphorylation, and ubiquitination (isopeptide bond formation). The protein contains several zinc-finger domains critical for DNA binding.
- GLI2 is a member of the GLI family of zinc finger proteins, implicated in developmental processes and linked to holoprosencephaly when mutated.
- Alternative splicing produces functionally distinct isoforms, with isoform 1 and 4 being detectable at the protein level.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 1100-1200 of human GLI2 (NP_005261.2). For Western blotting, a band of approximately 168 kDa is expected; consider using nuclear extracts and verifying with appropriate controls.
- For ELISA applications, the antibody may be used to detect peptide or recombinant protein; optimization of coating and detection conditions is recommended.
- Cross-reactivity is reported for human, mouse, and rat; validate specificity in the relevant sample system before use.
- As a polyclonal antibody, lot-to-lot variability is possible. We recommend careful titration and inclusion of positive/negative controls.
- Given GLI2’s involvement in the Hedgehog pathway, this antibody is suitable for studying transcriptional regulation in developmental and cancer models.
